Connect to VPC via VPN S2S links or Direct Link or VPC peering, or transit gateway. In enterprise cases, it is not a matter of "can stay within a private network", it is a must.

I do not think traffic between mule applications deployed to an Anypoint VPC and on-premises systems can stay within a private network. They would either go through IPSec tunnel, or via VPC attachment. depending on networking choosen.
