I am trying to understand the question correctly; it is not saying that they are using on-prem email exchange server, it can be o365 or gmail. hence if they need to deploy TE250X and would like to provide inline email security, I am thinking to have Harmony Email & Collaboration (HEC) and integrate HEC to TE250X. checking the network security deployment, all would require Security Gateway, even MTA also require Security Gateway. Because Stand alone is inline. Stand alone mainly is used when the GWs are from another vendor.

In-line mode is the same as Prevent mode. It always requires MTA, which means Security Gateway involvement.

Question asks: How would you deploy TE250X Check Point appliance just for email traffic and IN-LINE mode without a Check Point Security Gateway? The documentation that you've quoted said: Inline: This is a stand-alone option that deploys a TE250X inline OR on a SPAN port, and uses all of the Threat Prevention Blades. The question is talking about INLINE mode... your quote said that you can deploy INLINE ore a SPAN. Span is not inline. So, why should be A... and not B? It's clearly B.
