Question says MTU supported by VPN and DX which means via public VIF. MTU supported on this is 1500 only Private VIF and Transit VIF supports Jumbo frames

Dunno about VPN but since it travels on the Internet I would say No.
Direct Connect could support Jumbo frames. But when talking about VPN, if it's specified, it's always through Internet, so MTU 1500 is supported by all the Internet parties.
