Site to Site VPN with acceleration re-routes your data to AWS Global Acceleration endpoints first, then packets travel on AWS wires to its AWS destination, thus it's faster than traditional VPN connection via direct-connect. In order to do that, a transit gateway is a must.

Acceleration is only supported for Site-to-Site VPN connections that are attached to a transit gateway. Virtual private gateways do not support accelerated VPN connections. https://docs.aws.amazon.com/vpn/latest/s2svpn/accelerated-vpn.html
